The drug was used to treat cerebral circulatory insufficiency and brain arterial disease. The main active principle was isoxsuprine, largely used to treat cerebral and peripheral vascular diseases. In addition to isoxsuprine, the drug also contained (i) pyridoxine (vitamine B6) which is important for brain development, for nerve conduction and to reduce neuropathic pain; and (ii) α-tocopherol (vitamine E) which contributes to strengthen the body’s natural defense against infection. Vitamin E helps to boost the immune system. Isoxsuprine functions as a peripheral vasodilator, acting as a β2-adrenergic agonist and NR2B subtype-selective N-methyl-D-aspartate (NMDA) receptor antagonist. As such, it is a potent neuroprotective agent, considered useful for the treatment and prevention of ischemic stroke. In addition to its central effect, the drug directly affects the vascular smooth muscle, leading to a peripheral vascular dilation. This is why this oral drug is also used as a tocolytic agent, for treatment of pre-term labor and dysmenorrhea, via its capacity to relax the uterine smooth muscles. The drug has the capacity to slow or stop uterine contraction (tocolysis). Via its capacity to improve blood flow, the product can also contribute to protect from cardiotoxic damages. Isoxsuprine remains well used to day, both in human and veterinary medicines, to enhance blood flow and to improve maternal and fetal circulation. But the drug Trophodilan was essentially used to treat cerebrovascular insufficiency and mild traumatic brain injuries.
